CAR-modified marrow infiltrating lymphocytes efficiently target malignant plasma cells with very low antigen density

Researchers evaluated CAR-modified marrow infiltrating lymphocytes (CAR-MILs) for multiple myeloma therapy. Using super-resolution microscopy (dSTORM), they discovered that CAR-MILs efficiently eliminate autologous malignant plasma cells even at extremely low BCMA and TACI antigen densities. Standard flow cytometry significantly underestimated these levels. The findings demonstrate TACI's potential as a target and emphasize dSTORM's utility in assessing CAR sensitivity thresholds.
