Mezigdomide, carfilzomib, and dexamethasone versus carfilzomib and dexamethasone in patients with relapsed or refractory multiple myeloma (SUCCESSOR-2): a phase 3, open-label, randomised controlled trial
Last Updated: Tuesday, July 14, 2026
Researchers in the SUCCESSOR-2 phase 3 trial found that combining mezigdomide, carfilzomib, and dexamethasone significantly improved progression-free survival (18.0 vs. 8.3 months) in patients with relapsed or refractory multiple myeloma. Although the triplet therapy increased grade 3/4 adverse events, it remains a clinically meaningful option for predominantly triple-class-exposed, refractory patients with unmet needs.
