Mezigdomide, carfilzomib, and dexamethasone versus carfilzomib and dexamethasone in patients with relapsed or refractory multiple myeloma (SUCCESSOR-2): a phase 3, open-label, randomised controlled trial

The SUCCESSOR-2 trial demonstrates that adding mezigdomide to carfilzomib and dexamethasone significantly improves progression-free survival in patients with relapsed multiple myeloma. Median PFS reached 18.0 months versus 8.3 months. Despite higher rates of neutropenia and infections, this combination is a clinically meaningful option for anti-CD38 and lenalidomide-refractory patients.
