Clonal relationship and mutation analysis in lymphoplasmacytic lymphoma/Waldenström macroglobulinemia associated with diffuse large B-cell lymphoma

Researchers investigating the clonal relationship of lymphoplasmacytic lymphoma (LPL) and DLBCL together with mutations contributing to transformation among 13 patients with LPL/Waldenström macroglobulinemia who developed DLBCL found that although DLBCL presentations were clonally related to the dominant clone in LPL in the majority of patients, 23% of patients had DLBCL that was clonally unrelated to the major malignant B-cell clone in LPL. Further, 73% of patients with LPL displayed MYD88L265P mutation, while CXCR4 mutations were observed in 55%.
